چکیده

conclusions isaba1 has become an important factor in a. baumannii’s resistance to cefotaxime. results isaba1 located upstream of blaampc was found in 24 (48%) of the a. baumannii isolates. in all of the studied isolates that had isampc, the mic for cefotaxime was 64 - 256 μg/ml. based on the rep-pcr patterns among the resistant isolates, the highest number of isampc positive isolates belonged to type b (n = 19) and type c (n = 12). objectives we examined the prevalence isampc and its correlation with cefotaxime resistance. materials and methods standard biochemical tests were used to identify isolates. genomic species of the genus acinetobacter were confirmed by amplified ribosomal dna restriction analysis (ardra). the susceptibility of 50 a. baumannii isolates to a variety of antimicrobial agents was determined using the disk diffusion method and e-test strips. pcr was used to investigate the connection of insertion sequences and the ampc gene. clonal relatedness was determined by repetitive extragenic palindromic pcr. background infections due to acinetobacter baumannii have become a significant challenge in modern healthcare systems. the global upsurge of multidrug resistance in a. baumannii has created widespread problems in the treatment of patients.

ISAba1 targets a specific position upstream of the intrinsic ampC gene of Acinetobacter baumannii leading to cephalosporin resistance.

Sir, Resistance to third-generation cephalosporins such as ceftazidime and cefotaxime in Acinetobacter baumannii is often due to increased expression of an intrinsic ampC gene. Expression increases when the ISAba1 insertion sequence (IS) is present in the appropriate orientation upstream of the ampC gene, providing an outward-facing promoter that appears to be stronger than the intrinsic promot...

Tn6168, a transposon carrying an ISAba1-activated ampC gene and conferring cephalosporin resistance in Acinetobacter baumannii.

OBJECTIVES To explore the cause of third-generation cephalosporin resistance in Australian Acinetobacter baumannii isolates belonging to global clone 1 (GC1). METHODS GC1 isolates from Australia were tested for resistance to ceftazidime and cefotaxime using disc diffusion and MICs. PCR was used to determine the context of ISAba1-ampC configurations and amplicons were sequenced. The level of t...

Acinetobacter baumannii nosocomial infections.

UNLABELLED Nosocomial infections caused by strains Acinetobacter baumannii strands are a growing clinical problem. The occurrence of multidrug-resistant strands is observed and that limits the ways of therapy considerably. The aim of the study was to determine the rate of infection and susceptibility spectrum of the species Acinetobacter baumannii isolated from patients treated at Maria Skłodow...

Nosocomial Acinetobacter baumannii Infections and Changing Antibiotic Resistance

OBJECTIVES In the intensive care setting, Acinetobacter baumannii causes ventilator-associated pneumonia and other nosocomial infections that are difficult to treat. Objective of this study was to investigate nosocomial A. baumannii infections and its changing antibiotic resistance. METHODS A total of 56 patients diagnosed with A.baumannii infections between January 2009 and December 2011 wer...

Horizontal transfer of an ISAba125-activated ampC gene between Acinetobacter baumannii strains leading to cephalosporin resistance.

Sir, In Acinetobacter baumannii, resistance to third-generation cephalosporins such as ceftazidime and cefotaxime is known to arise as a consequence of acquisition of an insertion sequence, ISAba1, upstream of the chromosomal ampC gene. Indeed, ISAba1 is frequently found upstream of the ampC gene in isolates that are resistant to third-generation cephalosporins, and the promoter that directs tr...

in the GenBank non-redundant DNA database was examined. Two additional cases of ISAba1 associated with distinct ampC alleles were found in accession numbers EU604835 8 and AY325306. 3 In both of them, the IS was again in the same orientation and separated from the ATG initiation codon of ampC by 9 bp. The number of single nucleotide differences between the various ampC alleles is shown in Table...
